Physical Properties of Escherichia coli Spheroplast Membranes
AbstractWe investigated the physical properties of bacterial cytoplasmic membranes by applying the method of micropipette aspiration to Escherichia coli spheroplasts. We found that the properties of spheroplast membranes are significantly different from that of laboratory-prepared lipid vesicles or that of previously investigated animal cells. The spheroplasts can adjust their internal osmolality by increasing their volumes more than three times upon osmotic downshift. Until the spheroplasts are swollen to their volume limit, their membranes are tensionless. At constant external osmolality, aspiration increases the surface area of the membrane and creates tension. What distinguishes spheroplast membranes from lipid bilayers is that the area change of a spheroplast membrane by tension is a relaxation process. No such time dependence is observed in lipid bilayers. The equilibrium tension-area relation is reversible. The apparent area stretching moduli are several times smaller than that of stretching a lipid bilayer. We conclude that spheroplasts maintain a minimum surface area without tension by a membrane reservoir that removes the excessive membranes from the minimum surface area. Volume expansion eventually exhausts the membrane reservoir; then the membrane behaves like a lipid bilayer with a comparable stretching modulus. Interestingly, the membranes cease to refold when spheroplasts lost viability, implying that the membrane reservoir is metabolically maintained
Bridge scour evaluation based on ambient vibration
The vulnerability of bridges to hazards such as earthquakes, wind and floods necessitates special structural characteristics. To guarantee the stability of bridge structures, the precise evaluation of the scour depth of bridge foundation has recently become an important issue, as most of the unexpected damage to or collapse of bridges has been attributed to hydraulic issues. In this paper, a vibration-based bridge health monitoring system that utilizes only the response of superstructure to rapidly evaluate the embedded depth of a bridge column is proposed. To clarify the complex fluid-solid coupling phenomenon, the effects of embedded depth and water level were first verified through a series of static experiments. A confined finite element model simulated by soil spring effects was then established to illustrate the relationship between the fundamental frequency and the embedded depth. Using the proposed algorithm, the health of the bridge is able to be inferred by processing the ambient vibration response of the superstructure. To implement the proposed algorithm, a SHM prototype system monitoring environmental factors such as temperature, water level, and inclination was developed to support on-line processing. The performance of the proposed system was verified by a series of dynamic bridge scour experiments conducted in a laboratory flume and compared with readings from a water-proof camera. The results showed that using the proposed vibration-based bridge health monitoring system, the embedded depth of bridge column during complex scour processes is able to be reliably calculated
Energy and nitrogenous waste from glutamate/glutamine catabolism facilitates acute osmotic adjustment in non-neuroectodermal branchial cells
Maintenance of homeostasis is one of the most important physiological responses for animals upon osmotic perturbations. Ionocytes of branchial epithelia are the major cell types responsible for active ion transport, which is mediated by energy-consuming ion pumps (e.g., Na+-K+-ATPase, NKA) and secondary active transporters. Consequently, in addition to osmolyte adjustments, sufficient and immediate energy replenishment is essenttableial for acclimation to osmotic changes. In this study, we propose that glutamate/glutamine catabolism and trans-epithelial transport of nitrogenous waste may aid euryhaline teleosts Japanese medaka (Oryzias latipes) during acclimation to osmotic changes. Glutamate family amino acid contents in gills were increased by hyperosmotic challenge along an acclimation period of 72 hours. This change in amino acids was accompanied by a stimulation of putative glutamate/glutamine transporters (Eaats, Sat) and synthesis enzymes (Gls, Glul) that participate in regulating glutamate/glutamine cycling in branchial epithelia during acclimation to hyperosmotic conditions. In situ hybridization of glutaminase and glutamine synthetase in combination with immunocytochemistry demonstrate a partial colocalization of olgls1a and olgls2 but not olglul with Na+/K+-ATPase-rich ionocytes. Also for the glutamate and glutamine transporters colocalization with ionocytes was found for oleaat1, oleaat3, and olslc38a4, but not oleaat2. Morpholino knock-down of Sat decreased Na+ flux from the larval epithelium, demonstrating the importance of glutamate/glutamine transport in osmotic regulation. In addition to its role as an energy substrate, glutamate deamination produces NH4+, which may contribute to osmolyte production; genes encoding components of the urea production cycle, including carbamoyl phosphate synthetase (CPS) and ornithine transcarbamylase (OTC), were upregulated under hyperosmotic challenges. Based on these findings the present work demonstrates that the glutamate/glutamine cycle and subsequent transepithelial transport of nitrogenous waste in branchial epithelia represents an essential component for the maintenance of ionic homeostasis under a hyperosmotic challenge
THE EFFECT OF COUNTER MOVEMENT JUMP PERFORMANCE IN MIDDLEAGED ELDERLY PRACTICING TAI-CHI EXERCISE
The purpose of this study was to investigate biomechanical effects of Tal-Chi exercise on the lower-extremity in middle-aged elders during counter-movement jump. Twelve middle-aged elders with regular Tai Chi exercise experience and twelve healthy middle-aged elders participated in this study. Ten Vicon Motion System cameras, two Kistler force plates were used simultaneously to capture the kinematic and dynamlc parameters of standing vertical jumps. Independent samples &test was performed for statistical analysis ( u = .05 ). Since the jump height of Tai Chi group was significantly higher ( p c .05 ). It showed that practicing Tai Chi exercise could effectively slow down the degeneration of the moment and power at the hip Joint. Therefore, middle-aged elders were recommended to engage in long-term Tai Chi exercise
Resveratrol retards progression of diabetic nephropathy through modulations of oxidative stress, proinflammatory cytokines, and AMP-activated protein kinase
<p>Abstract</p> <p>Background</p> <p>Diabetic nephropathy (DN) has been recognized as the leading cause of end-stage renal disease. Resveratrol (RSV), a polyphenolic compound, has been indicated to possess an insulin-like property in diabetes. In the present study, we aimed to investigate the renoprotective effects of RSV and delineate its underlying mechanism in early-stage DN.</p> <p>Methods</p> <p>The protective effects of RSV on DN were evaluated in streptozotocin (STZ)-induced diabetic rats.</p> <p>Results</p> <p>The plasma glucose, creatinine, and blood urea nitrogen were significantly elevated in STZ-induced diabetic rats. RSV treatment markedly ameliorated hyperglycemia and renal dysfunction in STZ-induced diabetic rats. The diabetes-induced superoxide anion and protein carbonyl levels were also significantly attenuated in RSV-treated diabetic kidney. The AMPK protein phosphorylation and expression levels were remarkably reduced in diabetic renal tissues. In contrast, RSV treatment significantly rescued the AMPK protein expression and phosphorylation compared to non-treated diabetic group. Additionally, hyperglycemia markedly enhanced renal production of proinflammatory cytokine IL-1β. RSV reduced IL-1β but increased TNF-α and IL-6 levels in the diabetic kidneys.</p> <p>Conclusions</p> <p>Our findings suggest that RSV protects against oxidative stress, exhibits concurrent proinflammation and anti-inflammation, and up-regulates AMPK expression and activation, which may contribute to its beneficial effects on the early stage of DN.</p
Observational connection of non-thermal X-ray emission from pulsars with their timing properties and thermal emission
The origin and radiation mechanisms of high energy emissions from pulsars
have remained mysterious since their discovery. Here we report, based on a
sample of 68 pulsars, observational connection of non-thermal X-ray emissions
from pulsars with their timing properties and thermal emissions, which may
provide some constraints on theoretical modeling. Besides strong correlations
with the spin-down power and the magnetic field strength at the light
cylinder , the non-thermal X-ray luminosity in 0.5 - 8 keV, , represented by the power-law component in the spectral model, is found to
be strongly correlated with the highest possible electric field strength in the
polar gap, , of the pulsar. The spectral power index of that power-law component is also found, for the first time in the
literature, to strongly correlate with , and , thanks to the large sample. In addition, we found that can be
well described by , where
and are the surface temperature and the emitting-region radius of the
surface thermal emission, represented by the black-body component in the
spectral model. , on the other hand, can be well described only
when timing variables are included, and the relation is

between surface thermal emission and electron-positron pair production in

Psychometric evaluation of the Farsi version of the diabetes foot self-care bahavior scale
Background: Diabetes foot self-care is one of the self-management behaviors of diabetic patients leading to a reduction in the incidence of pressure ulcers and amputation. Having a valid, reliable, simple and comprehensive tool is essential in measuring the self-care behavior of diabetic patients. The aim of this study was to evaluate the psychometric properties of the Farsi version of the diabetes foot self-care bahavior scale (DFSBS) in Iran. Methods: In this cross-sectional and methodological study, 500 patients with type 2 diabetes were recruited by convenience sampling. Construct validity was assessed by exploratory factor analysis (over 300 patients) and confirmatory factor analysis (over 200 patients). Internal consistency was calculated by Cronbach’s alpha coefficient and its stability was calculated by intraclass correlation coefficient (ICC). Results: In the exploratory factor analysis, two self-care factors related to feet and shoes were extracted which had specific values of 38.49 and 1.24, respectively, and were able to account for 56.22% of the total self-care variance of diabetes foot. Confirmatory factor analysis had excellent fit model. The internal consistency and ICC of the whole instrument were 0.83 and 0.791 (95% CI: 0.575–0.925; P < 0.001), respectively. Conclusions: The Farsi version of DFSBS (F-DFSBS) has good validity and reliability, and due to its appropriate psychometric properties, this tool can be used in future studie
Image operator learning coupled with CNN classification and its application to staff line removal
Many image transformations can be modeled by image operators that are
characterized by pixel-wise local functions defined on a finite support window.
In image operator learning, these functions are estimated from training data
using machine learning techniques. Input size is usually a critical issue when
using learning algorithms, and it limits the size of practicable windows. We
propose the use of convolutional neural networks (CNNs) to overcome this
limitation. The problem of removing staff-lines in music score images is chosen
to evaluate the effects of window and convolutional mask sizes on the learned
image operator performance. Results show that the CNN based solution
outperforms previous ones obtained using conventional learning algorithms or
heuristic algorithms, indicating the potential of CNNs as base classifiers in
image operator learning. The implementations will be made available on the
